I do not care or know of the other 4 of the 5 choices for Spider-Man's reboot. SO I've put together whom I think could be in a realistic up coming reboot.
Peter Parker: They're considering Josh Hutcherson. He's of age now, only 18 (actually a teen year) GOOD actor, and fit. I think he'd be ideal given the other choices.
For his love interest Gwen Stacy, his first love, mind you from the comics, I choose Anna Sophia Robb. She is still in her teen years not far behind Josh, and a good actress. Plus she and Josh have worked together before in projects as child actors. So there's that familiarity.
For Eddie Brock who should be Peter's friend who is out of juvy, and looking to make things right, I choose Cameron Gignant. They both get jobs at the Bugle and their rivalry is friendly at first. (Don't make him Venom in the first movie, just get into his background, like how his parents were friends with Peter's and were scientists that died.)
Harry Osborn- Nicholas Purcell...
Uncle Ben- Richard Jenkins
For Aunt May- Jayne Atkinson
For Norman Osborn- Neal McDonough
OR Jason Isaacs
Doctor Octopus- Otto Octavious, head of Oscorp science staff to follow up Spectacular Spider-Man story arch- Michael Emerson
For the Lizard- Bruce Greenwood, I've always seen him as a Kurt Conners type myself
Now none of these foes have to (in fact they shouldn't) all become super villains at once. In fact, they should not be mutated in the first film.
Robbie Robbinson- Rondell Sheridan
Or Phill Morris
J. Jonah Jameson- Clancy Brown (Come on, tell me you can't imagine him screaming "PARKER!!")
For Captain Stacy- James Morrison
