I'll start off with the S.H.I.E.L.D. agents that we know of for sure, beginning with...
Nick Fury - Samuel L. Jackson (Iron Man, Iron Man 2)
The main man, Samuel L. Jackson was the muse for the Ultimates Nick Fury and in Iron Man he approaches Tony Stark for a project called the Avengers Initiative.
Phil Coulson - Clark Gregg (Iron Man, Iron Man 2, Thor)
Although Phil Coulson is not a character in the comics, he was the first agent of S.H.I.E.L.D. introduced into the Marvel movie universe and is played by Clark Gregg. He will be reprising his role in the upcoming Iron Man 2 and Thor.
Natasha Romanov AKA Black Widow - Scarlett Johansson (Iron Man 2)
Nobody knows what to think of this incarnation of the Black Widow. Will she be an American S.H.I.E.L.D. agent or a spy? My prediction is it will likely be something traitorous.

I read comic books on a daily basis and watch a ton of film and television, so I think I healthy understanding in what to look for when making one of these fan picks. Those are my credentials... I'm a media whore. So let's begin!
Timothy "Dum Dum" Dugan - Nick Offerman (Parks & Recreation)
I'd say he's the second most popular S.H.I.E.L.D. agent and is considered a "nasty old bastard" by Nick Fury.
I'm not a fan of Parks & Recreation, but I do like this actor.
Valentina la Fontaine AKA The Contessa - Monica Bellucci (The Sorcerer's Apprentice)
Just as deadly as any of the Howling Commandos, Nick Fury's ex and imported from Italy, Valentina should be younger than Fury, but only by a decade or two.
I've been a fan of Monica Bellucci after seeing her in The Matrix sequels and Le pacte des loups (Brotherhood of the Wolf). Since then, I have seen her in a string of awesome movies such as Shoot 'Em Up, Irréversible and Malèna. In my opinion, the sexiest and most talented actress ever.
Dino Manelli - Christopher Meloni (Oz, Law & Order SVU)
I like him in this role because I think he would make an interesting addition to the already epic cast I've got going on here.
I don't know anyone who doesn't love this guy! This is the only other actor I could see
replacing J. Jonah Jameson in the Spider-Man reboot, which is funny because Meloni starred in HBO's Oz series with J.K. Simmons.
Gabriel "Gabe" Jones - Taye Diggs (Private Practice)
This is another character who has a clean personality slate as far as comic book characters go. He'd have almost free reign to reinvent the character.
Taye Diggs is one of these under-rated actors who probably should be a bigger star. He's awesome in Equillibrium!
Maria Hill - Michelle Rodriguez (Lost, Avatar)
Honestly, I believe this is one of those instances where casting this person as this person just makes total sense and you can't even contest it! You can't!
Samuel "Snap" Wilson AKA The Falcon - Nelsan Ellis (True Blood)
I think the Falcon should look about Captain America(Chris Evans)'s age, so I nominate the hilariously freaky gay dude from True Blood.
Sharon Carter - Alice Eve (She's Out of My League)
Whoever lands the role of Sharon Carter should to be extremely attractive and needs to get in touch with their inner badass.
I don't know much about Alice Eve other than she's effin' hot as Hell and was up for the role of Peggy Carter in Captain America: First Avenger. The role ultimately went to Hayley Atwell.
Mitch Carson - Matt Dillon (Takers)
Half of Mitch Carson's face was burned off while attempting to stop another agent from fleeing the Helicarrier with a newly designed Ant-Man suit. The determination to bring the ex-agent to justice for mutilating his face is admired and could be a great character for an Ant-Man movie.
I'd like to see Matt Dillon in this role. I think he could bring the right kind of intensity and humour this storyline would need.
Eric O'Grady - Aaron Paul (Big Love, Breaking Bad)
An immoral agent steals the new Ant-Man suit from Henry Pym aboard the S.H.I.E.L.D. Helicarrier. If you haven't read it already, check out Robert Kirkman's short-lived The Irredeemable Ant-Man volumes and love it.
