“I do think it would be tricky to totally invent a major villain, but if I did that it would be The Mime. My big contribution would be a villain called The Mime. That’s a joke, by the way. A joke.”
Now as he said above, it's "a joke," but perhaps this casting call for clowns, magicians, and especially French mimes is to put an inside joke into the film. Just a thought.
Or, let's go really outside the box, and assume Marc Webb wants to explore the plot from the Marvel comic book,
Peter Parker: Spider-Man #38, which was, "Spidey faces his fiercest, and quietest, foes: murderous Mimes!" (Image on the right)

The Amazing Spider-Man 2 will be directed by Marc Webb from a script written by Alex Kurtzman, Roberto Orci, Jeff Pinkner and James Vanderbilt. The film will star Andrew Garfield as Peter Parker/Spider-Man, Emma Stone as Gwen Stacy, Sally Field as Aunt May, Shailene Woodley as Mary Jane Watson, Paul Giamatti as Rhino, Jamie Foxx is Electro and Chris Cooper as Norman Osborn. The Amazing Spider-Man sequel will swing into theaters May 2, 2014.
